Abstract

While the toxicity of many respirable particles has been well studied, much less is known regarding the potential adverse health effects of mineral clays even though human exposures are relatively widespread. This review was developed to improve understanding of the physical properties, interaction with biological materials, known human health effects, and laboratory studies (in vivo and in vitro) of a select set of mineral clays: montmorillonite, hectorite, and kaolinite. Review of literature suggests that even though large populations are often exposed to mineral clays there is evidence that prolonged exposures can produce acute and chronic pathologies in humans. Studies with animals support the notion that mineral clays induce respiratory toxicity and inflammation. In vitro studies with relevant cell models provide additional support that mineral clays can initiate inflammatory events. Taken together, additional research is needed to comprehensively understand adverse effects, the mechanisms involved and develop potential therapeutic interventions.
